Principle of the Assay::The Mouse CD22 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD22 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse CD22 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD22 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD22 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD22 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: CD22 or cluster of differentiation-22, is a molecule belonging to the SIGLEC family of lectins. This gene is mapped to 19q13.2. It is found on the surface of mature B cells and to a lesser extent on some immature B cells. Generally speaking, CD22 is a regulatory molecule that prevents the overactivation of the immune system and the development of autoimmune diseases. It is a negative regulator of antigen receptor signaling whose onset of expression at the mature B cell stage may serve to raise the antigen concentration threshold required for B cell triggering. CD22 functions as an inhibitory receptor for B cell receptor (BCR) signalling. This gene can downmodulate signaling through the IgM and IgD B-cell receptors(BCRs), but not through the IgG BCR, because the IgG cytoplasmic tail prevents CD22 phosphorylation and actually enhances IgG-BCR signaling
